وی پس از مدتی مدرك فوق لیسانس را اخذ و بدلیل علاقه و عشق به زبان و ادبیات فارسی، تصمیم گرفت تا از تمامی نقاط ایران همه اصطلاحات، لغات، امثال و حكم فارسی را جمع آوری و تدوین نماید.

وی در سال 1321 موفق به اخذ نخستین مدرك دكترای ادبیات و زبان فارسی از دانشگاه تهران شد و رساله خود را تحت عنوان 'مزدیسنا و تاثیر آن در ادبیات فارسی' با راهنمایی استاد ملك الشعرای بهار ارائه داد و در دانشگاه تهران مشغول به تدریس شد.

پس از انتقال این سازمان به دانشكده ادبیات دانشگاه تهران، ریاست آن طبق اساسنامه مصوب شورای دانشگاه، به عهده دكتر محمد معین گذاشته شد. در آن زمان وی هم زمان عضو شورای عالی فرهنگ بود.

استاد معین همچنین در كنگره جهانی ایران‌شناسان كه در سال1345 برگزار شد، ریاست كمیسیون ادبیات را عهده دار بود.

بنا به تصویب هیات وزیران وقت استاد معین به عضویت فرهنگستان ایران نایل آمد. او كه بیش از نیمی از عمر خود را صرف تحقیق در زمینه ادبیات فارسی كرده بود، نسبت به تدوین فرهنگ نامه ای كه به فرهنگ فارسی معین معروف است همت گماشت.

نگارش و تدوین حدود یك سوم جلد آخر این فرهنگ نامه، همراه با بیماری استاد شد، كه سید جعفر شهیدی، دوست و همكار استاد معین نسبت به اتمام آن اقدام كرد.

فرهنگ معین، فرهنگ نامه ای جامع در زبان فارسی است كه با توجه به هویت دستوری لغات و ریشه شناسی آنها، با متدی جدید تدوین شده است. این فرهنگ نامه شامل سه بخش لغات، تركیبات خارجی و اعلام است.
